ABSTRACT :

 

The basic number of chromosomes in the somatic cells of an individual or a species is called the somatic number and is designated 2n. In normal diploid organisms, autosomal chromosomes are presented in two copies. There may, or may not, be sex chromosomes. Polyploid cells have multiple copies of chromosomes and haploid cells have single copies. Thus, in humans 2n = 46. In the germ-line (the sex cells) the chromosome number is n (humans: n = 23). Identification of sex chromosome is a method that we can know what person he/she is. Normaly a man have chromosome XY dan female have XX but sometimes we have a problem to diagnosis. Is she with XX or XY? Or is he with XY or XX? To identify that we use karyotype method.

Karyotyping is process to make chromosome from  raw material ( whole blood/etc) until we get a chromosome graphic. Karyotypes describe the number of chromosomes, and what they look like under a light microscope. Attention is paid to their length, the position of the centromeres, banding pattern, any differences between the sex chromosomes, and any other physical characteristics. The preparation and study of karyotypes is part of cytogenetics. It took until the mid 1950s until it became generally accepted that the karyotype of humans included only 46 chromosomes.

Cytogenetics is a branch of genetics that is concerned with the study of the structure and function of the cell, especially the chromosomes. It includes routine analysis of G-Banded chromosomes and other cytogenetic banding techniques. Now the advance technique of molecular cytogenetics  are fluorescent in situ hybridization (FISH) and comparative genomic hybridization (CGH).
