M. leprae-derived Major Membrane Protein II (MMP-II) as A New Antigen for Serodiagnosis of Leprosy Infection

Leprosy infection is one of the most important health problems in Indonesia. Due to the lack of preventive measures, the number of new case remains the same level even after achieving the ‘Elimination Goal of WHO’ in 2000.
Development of sero-diagnostic techniques of leprosy is an important subject of study for control of leprosy. Because we can predict risky subclinical cases a few years prior to clinical manifestation.
M. leprae-derived major membrane protein, MMP-II, is a recombinant protein antigen established by one of the author (Y.M.). The serum IgG antibody titer of 353 various serum samples was tested by ABC-ELISA. 155 healthy Japanese sera were also tested as non-endemic control.
It was found that 6.3% of PB patients (1 out of 16), 52.6% of MB patients (30 out of 57), 54.5% of RFT (36 out of 66), 50% of health person in East Jawa (43 out of 86), 8.1% of Household contacts (7 out of 86) and 14.3% of Neighbor contact (6 out of 42) was positive. The highest average antibody titer was 0.943 OD units in MB and followed by RFT (0.638) and endemic control (0.353).
The results of detailed analysis of serological characteristics of MMP-II will be discussed in the presentation.
